Description
English: Lineage tracing of Ret -expressing ureteric bud tip cells in renal organ culture. A Ret-CreERT2/+; Rosa26R-tdTomato/+; Hoxb7-myrVenus/+ kidney was excised at E12.5, treated for 1 h with 1 μm 4-OH tamoxifen, washed several times with PBS and cultured in normal medium. Images were collected every 20 min. The image on the left shows the green channel (showing the entire ureteric bud epithelium) and the red channel (showing tdTomato+ cells), while the image on the right shows only the red channel. At ~10 h, red cells start to appear in the extreme tips of the ureteric bud. As the tips extend and branch, the tips remain tdTomato+, as do the newly formed trunks (indicated by arrows in last frame). The yellow numbers at bottom left show the elapsed time since the start of the movie (h:min:s on day 1 and d:h:min:s on days 2–3); there is a gap in the movie between 21h:22 min and 1d:1h:9min.
